چکیده
Oxidative stress plays an important role in the pathogenesis of heart disease. Tocotrienol-rich fraction (TRF) is antioxidant and that has potential to reduce risk This study determine protective effects palm TRF against H2O2-induced oxidative damage neonatal rat cardiomyocytes (NRCM). The NRCM were divided into control, treated with (10 µg/mL), H2O2 (0.5 mM) prior induction (pre-treatment). Cell viability was determined by MTS assay, while presence reactive oxygen species (ROS) using fluorescent dihydroethidium (DHE) 5-(and-6)-carboxy-2′,7′-dichlorodihydrofluorescein diacetate (carboxy-H2DCFDA) dye. Mitochondrial integrity cell death JC-1 Annexin V-FITC staining, respectively. Lactate dehydrogenase (LDH) superoxide dismutase (SOD) activity colorimetric assay kit. concentration from 0.5 5 mM reduced IC50 value used experiment. increased intensity carboxy-H2DCFDA DHE-stains; also green fluorescence J-monomers staining compared control group. LDH while SOD decreased Pre-treatment intensities DHE-stains, as well JC-1. Meanwhile, pre-treatment group but no changes recorded Palm protects reducing ROS production maintaining mitochondrial membrane thus death.

Palm tocotrienol-rich fraction supplementation suppressed stress-induced gastric oxidative stress in rats
Palm tocotrienol-rich fraction (TRF), an extract from palm oil containing both tocotrienol and tocopherol, is known for its antioxidant effect. The present study investigated the effect of palm TRF on the stomach using a water immersion restraint stress (WRS) model. Fortytwo male Sprague-Dawley rats were divided into three groups and were administered 60 mg/kg/d of palm TRF, α-tocopherol or veh...

متن کاملLevocarnitine Protects H9c2 Rat Cardiomyocytes from H2O2-induced Mitochondrial Dysfunction and Apoptosis
BACKGROUND Although the protective effects of levocarnitine in patients with ischemic heart disease are related to the attenuation of oxidative stress injury, the exact mechanisms involved have yet to be fully understood. Our aim was to investigate the potential protective effects of levocarnitine pretreatment against oxidative stress in rat H9c2 cardiomyocytes. METHODS Cardiomyocytes were ex...

In the present study, tocotrienol rich fraction (TRF) nanoemulsions were produced as an alternative approach to improve solubility and absorption of tocotrienols. In the present study, droplet size obtained after 10 cycles of homogenization with increasing pressure was found to decrease from 120 to 65.1 nm. Nanoemulsions stabilized with Tween series alone or emulsifier blend Brij 35:Span 80 (0....
متن کاملDownregulation of microRNA-100 protects H2O2-induced apoptosis in neonatal cardiomyocytes.
Hypoxia or reoxygenation-induced cardiomyocyte apoptosis is one of the major causes of cardiac dysfunction. Recently, regulations of microRNAs were shown to play important roles in cardiomyocyte apoptosis. MicroRNA-100 (miR-100) is one of the cardiac miRNA that was up-regulated in failing heart. In this study, we identified that miR-100 expression was up-regulated in H2O2-induced apoptosis in n...
